Somatic embryogenesis represents a valuable tool for the studies on the basic aspects of plant embryo development. Today this process is used as a potencial technique for large-scale plant micropropagation although, so far, it has been applied to only a small number of species. However, when somatic embryos are malformed they are considered economically useless. In Acca sellowiana (O. Berg) Burret, an important fruit-producing crop, large amounts of anomalous somatic embryos (76.3%) were found just after 40 days of culture of explants in a 2,4-D containing medium. Inoculation with arbuscular mycorrhizal fungi (AMF) of tree seedlings in the nursery is a biotechnological strategy to improve growth, survival after transplanting, biomass production and to reduce the use of fertilizers. Archontophoenix alexandrae and Euterpe edulis are palm species used in southern Brazil to produce the palm heart, the latter being included in the list of threatened species due to the overexploitation of its native population. The rhizomes of Zingiber officinale Roscoe (ginger) are widely used for their medicinal and flavoring properties, whereas the influence of root symbionts on their growth is poorly understood. In this study, the effects of phosphate fertilization and inoculation with a mixture of arbuscular mycorrhizal fungi (AMF) (isolates Glomus clarum RGS101A, Entrophospora colombiana SCT115A and Acaulospora koskei SPL102A) on survival, growth and development of micropropagated ginger were investigated. No Laboratório de Fisiologia do Desenvolvimento e Genética Vegetal do CCA-UFSC, nos anos de 1993 a 1996, foram testadas 24 combinações de tratamentos envolvendo dois genótipos de abacaxizeiro (Ananas comosus (L.) Merr.), seis combinações dos fitorreguladores ácido naftalenoacético (ANA) e 6-benzilaminopurina (BAP), e os meios de cultura líquidos e geleificados, com o objetivo de estabelecer um protocolo regenerativo para a micropropagação do abacaxizeiro. A taxa de regeneração comportou-se de forma quadrática para a maioria das combinações testadas. A Acca sellowiana Berg. (Myrtaceae) é uma frutífera nativa dos planaltos meridionais do Sul do Brasil e que se encontra em processo de domesticação. Seus frutos são doce-acidulados e podem ser consumidos in natura ou empregados para a produção de sucos e doces. Assim, informações sobre o desenvolvimento, morfologia e anatomia de seus frutos são de grande interesse e foram objetos do presente trabalho. O trabalho objetivou avaliar a fertilidade e as reservas de C e N em gemas de ramos, na entrada da dormência e dormência plena, das variedades 'Cabernet Sauvignon' e 'Nebbiolo'. Em um vinhedo de 'Cabernet Sauvignon' e outro de 'Nebbiolo', em São Joaquim (SC), foram coletadas gemas em ramos de ano, em duas épocas: na entrada da dormência (maio de 2012) e na dormência plena (agosto de 2012). Os tratamentos foram: (T1) gemas basais (1ª a 5ª); (T2) gemas medianas (6ª a 10ª), e (T3) gemas distais (11ª a 15ª). ABSTRACT Although karyotype features are useful data for evolutionary studies, cytogenetic data in Bambusoideae are mainly based only on chromosome counts. The first comparative cytogenetic analysis of three Neotropical woody bamboo species - Guadua chacoensis, G. angustifolia and Chusquea tenella - was undertaken based on new and reviewed chromosome counts, CMA/DAPI double staining, fluorescent in situ hybridization (FISH) with 35S and 5S rDNA probes, and genome size estimation. The two species of Guadua were found to have 2n = 46 chromosomes, while the first record for C. tenella was 2n = 44. The outer and inner integuments are double-layered except in the micropyle, where they are composed of more layers; the micropyle is zig-zag shaped. The egg apparatus lies at the micropylar pole, and the zynergids present a conspicuous filiform apparatus. We have investigated the effects of phosphate fertilization and inoculation with isolates of arbuscular mycorrhizal fungi Scutellospora heterogama SCT120E, Gigaspora decipiens SCT304A, Acaulospora koskei SCT400A, Entrophospora colombiana SCT115, and an assemblage (Mix) of all four isolates on growth, development and oleoresin production of micropropagated Zingiber officinale. After 120 and 210 d of growth, the Mix and phosphorus addition significantly increased shoot height relative to control plants. The γ-aminobutyric acid (Gaba) is a non-protein amino acid found in prokaryotes and eukaryotes. Its role in plant development has not been fully established. This study reports a quantification of the levels of endogenous Gaba, as well as investigation of its role in different stages of somatic embryogenesis in Acca sellowiana Berg. (Myrtaceae). Zygotic embryos were used as explants and they were inoculated into the culture medium contained different concentrations of Gaba (0,2, 4, 6, 8 and 10 µM).
